When I last used the Linux port of the AGS interpreter some time ago it was able to use the information in the exe to play the game without the need for a.dat. I don't see why this behaviour would have changed.
No you won't be able to run this under ScummVM, it uses the closed source Adventure Game Studio system not SCUMM/SPUTM or any other engine we support.
That said there is a Linux port or the AGS interpreter available, although going on what others have said I believe that doesn't work with this remake for whatever reason.
The version of Sam & Max will more than likely be the updated directx version by by Aaron Giles of MAME and ex-LucasArts fame.
Previously this updated binary (resources are the same I believe) was only available in parts of the UK and Europe in a compilation pack know as the Entertainment Pack. As LucasArts don't have this available for people who already have the game, preordering Armed & Dangerous to get the bonus disc is the only way many people can get an updated version from LucasArts, or your could just look here.
Yes, its also a lot of directx code to sort through for people who aren't familiar with directx. If anyone wants to help out grab CVS or a source code snapshot and look in the bs2 directory. All the rendering functions and a few other things are currently stubbed out. Patches are very welcome:)
The playable demo should work as well as the game for testing things out.

This talk should be similar to the one given on Sunday at 11 am at the Australian Game Developers Conference http://www.agdc.com.au/sched.php.
